Answer
The biotinylation of ab20640 was accomplished by covalently coupling the primary antibody free amine groups with the N-Hydroxysuccinimide ester of biotin under mild conditions to give a high degree of substitution.

Question
I did make an attempt to do an Abreview on ab68451. The application I used the antibody for was not listed, and since I couldn't successfully use it in the assay, it was impossible to provide any information. Therefore, I like to ask you let me have ab137967. Hopefully it will work for me in the assay and I'll be able to share the results. Thank you for your understanding. Best regards,

Answer
Thank you very much for your reply.
I'm sending a free of charge vial of ab137967 on the order ***. This antibody is back-ordered until September 10, and I expect it to arrive shortly after that date.
Please note that as we have not tested this antibody in your assay, we can not guarantee it to work, though I am very hopeful it will work better than ab68451. Please keep me updated about the results, and let me know if there is anything else that we can do for you.
Have a nice day!

Answer
Thank you for contacting Abcam earlier today.
I have talked to the lab about your enquiry regarding ab20640 and they have informed me that the antibody has been tested and is reactive with cell surface antigens on intact E. coli organisms. The immunogen is a mixture of whole cells from several E.coli serotypes, but some of the organisms in the immunogen may become disrupted during the immunization process. The antibody is a polyclonal antibody and will therefore recognize multiple epitopes on multiple proteins. Some of the epitopes recognized may be on E. coli proteins that are not cell surface proteins, but this has not been tested.
Please let me know if there is anything else I can help you with.

Answer
Thank you for contacting us. Unfortunately, the biotin/antibody ration is not determined for this antibody. As I mentioned, a common estimate for antibody biotinylation in general is is 3 - 6 biotin molecules per immunoglobulin. Please let us know if you need any other information.

Question
Our customer is looking for E. coli BL21 strain biotinylated antibody. Kindly advise if it will be possible to perform biotinylation of ab25823 with EasyLink kit and to leave the antibody active in ELISA. Is there any chance that ab20640 will recognize BL21 (sorry, but, unfortunately, I can't cross the info about Bl21 and the serotypes)? Thank you very much for your help.

Answer
Thank you for contacting Abcam.
For ab25823, it should be fine to conjugate the antibody with biotin, using one of our EasyLink kits.
As for ab20640, it may react with BL21, but I cannot guarantee it, all we know is that it will recognize the strains O18, O20, O44, O55, O111, O112, O125, O157, K12.
Please let me know if there is anything else I can help you with.

Question
We just bought a biotin tagged anti E. coli antibody (ab20640) and anAnti-E. coli LPS antibody [2D7/1] ( ab35654), I want to ab20640 conjugated with streptavidin-HRP, and ab35654 got biotinylated, soI am wondering the molecular weight of these antibody, and the molar concentration. Thank you very much.

Answer
Thank you for your enquiry.Unfortunately, it is not known exactly how much biotin per molecule of antibody is conjugated, the molecular weight of ab20640 is therefore undetermined. For ab35654, which is an unconjugated immunogloblulin, the molecular weight is ~150 kDa.As shown on the product datasheet, the concentration of ab20640 is 4 mg/ml and the concentration of ab35654 is 0.5 mg/ml.I hope this information is nevertheless helpful to you. Please do not hesitate to contact us if you need any more advice or information.

Question
I tried to bind E.coli K12 and strepvidin beads via ab20640. However, I did not get good results. Acording to description of ab20640, I thought it can react with K12. But Acording to another enquire of this produce, it seems that ab20640 is not against K12. I'm a little confused. Can ab20640 react with E.coli K12 in fact? If the answer is yes, Could you kindly provide some suggestions that can help me imrove the binding experiment?

Answer
Thank you for your enquiry. I can confirm that ab20640 Anti-E. coli antibody (Biotin) will recognize the K12 strain of E. coli. This antibody has been tested in ELISA against E.coli serotypes O157:H7, O20, O55, O111, O125 and K12. It will therefore be suitable for detection of the K12 strain. I hope this will be helpful to you. Should you have any further questions, please do not hesitate to contact us.
